Covaxin wiki: covid-19 vaccine manufactured by the Bharat biotech with the alliance of- Indian council of medical research and the national institute of virology is said to be India’s indigenous covid vaccine. The inactive virus-based vaccine is developed in Bharat Biotech’s BSL-3 with high containment facility.
Why use an inactivated vaccine?
Well, developing inactivated vaccines is a conventional method and has been used for decades. Even several other vaccines like influenza, polio, rabies, pertussis, all used the same technology for developing vaccines. The method can deliver more than 300 million doses, hence it is a well-established and safe platform in the world of vaccine technology.
Phase 1 and 2 trials
The virus strains were approved by ICMR and the national institute of virology in may 2020, to develop a complete indigenous covid-19 vaccine.
Covaxin entered the human trial in July after it was found safe on animals in the preclinical trial. India’s first indigenous vaccine was tried in the age group of 18-55 years with no case comorbidity. Those who had some comorbidities were taken out of the trial including pregnant women. Although, the volunteers were analyzed for hypertension, diabetes, kidney function, and immunity. The healthy volunteers who cleared the test were allowed for the trial.
However, in June 2020 the company got permission for phase 1 and phase 2 trials on humans for a developmental covid-19 vaccine, referred to as covaxin by the drug controller general of India, and the government
of India. The randomized trials consisted of double-blind and placebo-controlled methods, for which almost 12 sites were selected by ICMR.
The results of the phase 1 trial were announced by the company in December 2020 and put forward results through medRxiv preprint. The report was then published in “the lancet”.
Result of phase 2 trial
Again the phase 2 trial’s result was published in the lancet, in march 2021. The study represented that the phase 2 trial was more efficient with higher immune response and increased response of T-cell due to the difference of four weeks between phase 1 and phase 2 vaccine doses. The second trial on the same volunteers was done after four weeks of the previous trial, which showed the higher efficacy of the vaccine after the second dose or phase 2.
Phase 2 was started in September 2020, when phase 1 was about to analyze the safety of vaccines, phase 2 was about to analyze the efficacy and response of the vaccine.
After the success of phase 1 and phase 2, covaxin got permission for the third trial on humans. The trials were performed over the age group of 18 and above. This trial also included the double-blind and placebo-controlled study among 18 and above volunteers. Phase 3 of covaxin started on 25 November 2020. The third trial included about 26,000 volunteers from all over the world. For phase 3, about 22 sites were selected consisting of several states in the whole country such as Karnataka, Delhi, and West Bengal.
The efficacy reported after phase 3 was 81%, which then dropped to 78% in April 2021, reported by Bharat biotech.

The company fixed two different prices for state government and private hospitals, followed by 600 for state government and 1200 for private hospitals. However, covaxin price is higher than that of covidshield developed by the serum institute of India.
Covaxin side effects
Nonetheless, covaxin has some mild side effects similar to the covid infection. Although, no medical treatment is required for these mild effects. A little fever after is considered a healthy response, as when your immune system responds to some foreign material, it causes your body to produce certain changes to which your body is reacting. Not necessarily everybody will experience it, but still, a large number of people do feel some mild changes.
The common covaxin side effects include:-

Dosage of covaxin
Covaxin is given in two doses, as permitted by the drug controller general. The second dose should be given after 28 days of the first dose. As there should be a gap of 4 weeks between the two doses for maximum efficacy.
How does covaxin work?
The inactivated virus does not replicate inside the human body. It only instructs the immune system to fight against the virus and induces antibodies. The partially killed pathogen when enters the body, is recognized as a foreign particle and the body fights with it by producing antibodies to tackle the further encounter. The whole virion technology is used earlier for several vaccines including influenza vaccine, polio vaccine, rabies, and many others.
